- GOBLET Cohort 4 evaluated pelareorep with atezolizumab in second-line or later squamous cell anal cancer.
- Median overall survival had not yet been reached, leaving longer follow-up important for the registration case.
- Oncolytics is considering further studies and a possible accelerated approval pathway, subject to FDA agreement.
Oncolytics Biotech reported updated clinical findings on October 27, 2026, from Cohort 4 of its Phase 1/2 GOBLET study in previously treated squamous cell anal cancer. The company said the results support further development of pelareorep with the checkpoint inhibitor atezolizumab. The announcement was carried by GlobeNewswire.
Clinical signal supports next steps
The median duration of response was approximately 15 months, compared with a historical 9.5-month result for retifanlimab monotherapy in a similar patient population. The comparison was not based on a head-to-head trial. Median overall survival had not yet been reached, while all responding patients survived beyond 12 months and several survived beyond 24 months.
Oncolytics said the findings could support an accelerated approval pathway, although that route remains subject to agreement with the U.S. Food and Drug Administration. The company plans to explore additional clinical studies with a potential checkpoint-inhibitor partner to support registrational development in anal cancer.
Pelareorep targets gastrointestinal cancers
GOBLET is being conducted across 17 centers in Germany and covers several gastrointestinal cancer settings. Cohort 4 combines pelareorep with atezolizumab in second-line advanced and unresectable anal cancer. Oncolytics is also pursuing pelareorep development in colorectal and pancreatic cancer, according to its investor overview.
Company focuses on immunotherapy
Oncolytics Biotech is a Canada-based clinical-stage biotechnology company developing pelareorep, an intravenously delivered investigational immunotherapy. Its main focus is on gastrointestinal cancers, including colorectal, anal and pancreatic cancer, with development programs built around chemotherapy and checkpoint-inhibitor combinations. The company remains a specialized clinical developer rather than a commercial oncology manufacturer. Its approach is centered on using pelareorep to stimulate immune responses against tumors.
