The euphoria that propelled Spyre Therapeutics to a fresh 52-week high evaporated swiftly this week. After touching 90.00 euros on Tuesday, the stock suffered a sharp reversal, tumbling 7.1 percent to 78.50 euros on Wednesday — and later sliding further to 77.00 euros, a full 8.88 percent loss from the previous session. The pullback came as director Michael Thomas Henderson unloaded 20,000 shares at $100 each through a pre-arranged 10b5-1 trading plan that was set up back in February, a detail that tempers any suggestion of an opportunistic exit.

The selloff, however, runs counter to a recent wave of clinical optimism. On June 15, Spyre reported positive 12-week data from its Phase 2 SKYLINE trial, in which the candidate SPY002 met the primary endpoint in ulcerative colitis. The RHI score — a measure of intestinal inflammation — showed a statistically significant improvement. That result, combined with a safety signal from a competitor that has given the entire TL1A class a tailwind, triggered a flurry of analyst upgrades.

Deutsche Bank responded by lifting its price target on the stock from $115 to $135 while maintaining its buy rating. Wedbush followed with an increase to $100, and BTIG raised its target to $112. In each case, analysts cited a higher probability of success for the company's TL1A pipeline, which includes both SPY002 and the next-generation candidate SPY003.

Should investors sell immediately? Or is it worth buying Spyre Therapeutics?

The company is now advancing multiple late-stage programs. For SPY002, the next milestone is topline data from the SKYWAY study in rheumatoid arthritis, expected in the third quarter. Results for psoriatic arthritis and axial spondyloarthritis are slated for the fourth quarter. Separately, Spyre's SPY003 program — which targets a longer-acting antibody for inflammatory bowel disease — is on track to deliver monotherapy data in ulcerative colitis by mid-2026, with additional readouts across other indications, including rheumatoid arthritis, in the fourth quarter of that year.

Financially, Spyre sits on a debt-free balance sheet, bolstered by a recent capital raise that secured funding for research and manufacturing. Yet the heavy spending required to run multiple Phase 2 studies simultaneously means the company may need to tap the markets again before any revenue materialises — a familiar pattern for development-stage biotechs.

The insider sale, though modest in scale relative to the company's overall float, has added a layer of unease to an otherwise bullish narrative. Still, the fact that it was executed under a pre-planned trading mechanism suggests it was not a reactive move to the recent rally. The more important question for investors is whether the upcoming clinical milestones can sustain the momentum needed to push the stock back above the 90-euro threshold — and whether the market's current appetite for risk in the biotech sector will hold.
