BioNTech's oncology pipeline delivered two readouts within days of each other this month, and the contrast between them is hard to miss. One program, partnered with Bristol Myers Squibb, put up numbers the companies could present without caveats. The other, developed with China's DualityBio, cleared its primary endpoint but left the partners facing an awkward conversation about what comes next.

The stronger of the two came in lung cancer. On October 25, BioNTech and Bristol Myers Squibb unveiled Phase 2 data from the ROSETTA Lung-107 trial, testing pumitamig in combination with docetaxel in metastatic non-small cell lung cancer. The confirmed objective response rate landed at 36.7%, according to the companies. Median progression-free survival came in at 6.9 months. Every patient in the study had already received first-line immunotherapy plus chemotherapy before enrolling, meaning the results reflect the combination's performance specifically in the post-treatment setting.

A Breast Cancer Win That Raises Questions

Roughly a week earlier, BioNTech and DualityBio disclosed top-line results from the Phase 3 DYNASTY-Breast02 study of trastuzumab pamirtecan. The trial met its primary endpoint on progression-free survival versus physician's choice of chemotherapy in patients with HR-positive, HER2-low metastatic breast cancer. The improvement was statistically significant. It was also modest — the companies themselves characterized the potential benefit to patients as limited. Overall survival data were not yet mature.

Should investors sell immediately? Or is it worth buying BioNTech?

Then came the safety signal. Interstitial lung disease appeared more frequently than in earlier studies, with a small proportion of events graded 3 to 5. Reuters reported that this included severe and fatal cases. The disclosure, made on October 26, came without a decision on how the program will proceed. BioNTech and DualityBio said they will review the findings and determine next steps, leaving the future of trastuzumab pamirtecan genuinely open.

Gilead Deal Extends the Pumitamig Story

Pumitamig's reach is not confined to lung cancer. About two weeks ago, BioNTech and Gilead Sciences agreed to a clinical collaboration covering both development and supply. The plan calls for a Phase 3 study pairing pumitamig with Gilead's sacituzumab govitecan — marketed as Trodelvy — in first-line treatment of locally advanced or metastatic triple-negative breast cancer with low or absent PD-L1 expression. Under the arrangement reported by Reuters, BioNTech will sponsor and lead the trial while Gilead supplies the drug. The planned study amendments still require regulatory clearance.

Melbourne Plant and a Quarterly Date

Manufacturing capacity is expanding in parallel. Media reports indicate BioNTech opened an mRNA research and clinical-scale production facility at La Trobe University in Melbourne, Australia, roughly two weeks ago.
