BioNTech's Pipeline Math: One Analyst's $138 Target, Seven Pivotal Trials, and a Roche Handoff
Published on 11/04/2026 at 11:10 | Editorial boerse-global.deJefferies keeps its Buy rating and $138 price target on BioNTech, but the note's real weight sits on a single oncology asset — Pumitamig — as the company spreads its clinical and financial bets across partners.
Analyst Akash Tewari's report, published yesterday, opens with the familiar comfort of a beat: revenue and earnings per share both came in ahead of consensus. What follows is less comfortable. The quarterly print, in his view, is not what will move the stock. Pumitamig is.
The scale of the bet is visible in the study plan. BioNTech intends to run seven global pivotal trials with the candidate through the end of 2026, a schedule that leaves little doubt about where the oncology strategy is anchored. For shareholders, that shifts the relevant question from last quarter's numbers to the readouts still ahead.
Early response rates sketch the opportunity
Some of the first evidence comes from combining Pumitamig with a B7-H3 antibody-drug conjugate in small-cell lung cancer, where results split sharply by treatment line. The objective response rate reached 92% in the first line, 76% in the second, and 52% from the third line onward. Those are clinical response figures — not sales, and not yet a proven commercial outcome.
Should investors sell immediately? Or is it worth buying BioNTech?
Gotistobart, another core oncology program, has produced its own datapoint in squamous lung cancer: a median overall survival of 18.5 months against 10 months, with a hazard ratio of 0.56. A first interim analysis for that program is expected before the end of 2026, giving investors a second clinical marker alongside the expanding Pumitamig schedule.
The pipeline is not uniformly promising. Two mRNA cancer studies missed their goals or were discontinued — a reminder that progress on one candidate cannot be extrapolated across the portfolio, and that a blanket endorsement of the whole pipeline would be premature.
Roche takes the cost line from 2027
Financing all of this is less of a strain than it once was. BioNTech ended the third quarter of 2026 with cash and marketable securities of EUR 16.2 billion, a cushion that covers the research bill but says nothing about whether the research will pay off.
That cushion is also being shared. BioNTech disclosed yesterday that it has exercised an option in its collaboration with Roche and Genentech, shifting the program's future research and development costs principally to the two partners from January 1, 2027. BioNTech stands to collect royalties if development and commercialization succeed — a payout still tied to outcomes it does not fully control. Guidance for its own adjusted R&D and adjusted sales and administrative expenses was left unchanged.
A quarter of two halves
The financial backdrop is more mixed than a single quarter suggests. Revenue for the third quarter of 2026 came in at EUR 1,289.2 million, with IFRS net profit of EUR 184.5 million. Across the first nine months, however, BioNTech booked revenue of EUR 1,512.9 million against a net loss of EUR 1,168.2 million.
BioNTech at a turning point? This analysis reveals what investors need to know now.
A third alliance landed on October 22, when BioNTech and Gilead Sciences announced a clinical collaboration covering the supply of study drugs to test Pumitamig in combination with Gilead's sacituzumab govitecan. The vehicle is the global Phase 3 ROSETTA Breast-01 trial, subject to regulatory approval, in patients with previously untreated, locally advanced or metastatic triple-negative breast cancer whose tumors show low or no PD-L1 expression.
Not every development is external. According to a dpa-AFX report from yesterday, BioNTech's works council has criticized management's treatment of employees in a letter to the executive board — an internal friction point running alongside the company's transformation.
Jefferies' recommendation, then, pairs a solid set of numbers with a clear clinical condition: Pumitamig has to back up expectations with further credible data. Paying for the development and succeeding at it remain two separate questions.
